Steven Soderberghin draamassa lahjakas kirurgi John Thackery koettelee lääketieteen, moraalin ja rotunäkemysten rajoja 1900-luvun New Yorkin keskustasairaalassa. Knick-sairaalan vauraat tukijat häipyvät, köyhät potilaat lisääntyvät ja talous on retuperällä, mikä mullistaa sairaalan arjen. Thackery on yksi sairaalaan jäävistä sankareista ja yrittää selvittää liutaa lääketieteellisiä mysteerejä kamppaillen samalla kokaiiniriippuvuuden kourissa. Lopulta vastahakoinen Thackery saa työparikseen nuoren Algernon Edwardsin, jonka äly ja sumeilemattomat keinot vetävät vertoja hänen omilleen ja joka herättää niin lääkärikollegoiden halveksunnan kuin potilaiden epäluulotkin.
